We are planning on painting woodwork in the house. Do I need to paint all rooms? Can I leave one room, great room, with it’s stain finish?

Yes you can. The finish, paint or stain, is mostly decorative. If stained wood goes better with your great room decor then I don't think this would be a problem. If it looks good to you then it is the right choice.
