It will look fine as long as the flat paint is well painted, no thick roller marks, etc. Actually, with some dark or dramatic colors starting with a coat or two of flat is the only way to achieve complete coverage without the background showing through.

In addition if you have any Spackled repair areas, you need to spot prime them first, otherwise they will appear dull under a semi finish… use a Pigmented Shellac Sealer Primer..
